AP2M1 Antibody (SD0746)

Western Blot: AP2M1 Antibody (SD0746) [NBP2-67762] - Analysis of AP2M1 on PC-3M cell lysates. Proteins were transferred to a PVDF membrane and blocked with 5% BSA in PBS for 1 hour at room temperature. The primary antibody (1/500) was used in 5% BSA at room temperature for 2 hours. Goat Anti-Rabbit IgG - HRP Secondary Antibody at 1:5,000 dilution was used for 1 hour at room temperature.

Immunocytochemistry/Immunofluorescence: AP2M1 Antibody (SD0746) [NBP2-67762] - Staining AP2M1 in MCF-7 cells (green). The nuclear counter stain is DAPI (blue). Cells were fixed in paraformaldehyde, permeabilised with 0.25% Triton X100/PBS.

Immunohistochemistry-Paraffin: AP2M1 Antibody (SD0746) [NBP2-67762] - Analysis of paraffin-embedded mouse brain tissue using anti-AP2M1 antibody. Counter stained with hematoxylin.

Flow Cytometry: AP2M1 Antibody (SD0746) [NBP2-67762] - Analysis of AP2M1 was done on PANC-1 cells. The cells were fixed, permeabilized and stained with the primary antibody (1/50) (red). After incubation of the primary antibody at room temperature for an hour, the cells were stained with a Alexa Fluor 488-conjugated Goat anti-Rabbit IgG Secondary antibody at 1/1000 dilution for 30 minutes.Unlabelled sample was used as a control (cells without incubation with primary antibody; black).

Background

AP2M1 is a subunit of the heterotetrameric coat assembly protein complex 2 (AP2), which belongs to the adaptor complexes medium subunits family. It is required for the activity of a vacuolar ATPase, which is responsible for proton pumping occurring in the acidification of endosomes and lysosomes. It may also play an important role in regulating the intracellular trafficking and function of CTLA-4 protein.
