Description | Calcein AM is a viability dye that can be used in permeabilized cells. Upon uptake by a live cell, Calcein AM is cleaved by intracellular esterases resulting in fluorescence. Dead cells will not have sufficient esterase activity and will not fluoresce. In this way, dead cells may be identified by their low Calcein signal. Calcein is efficiently excited by the 488nm blue laser and emits at 516 nm (FL1 on a standard 4 color flow cytometer). |

Application Notes | Calcein AM should be used at as low a final working concentrations as possible to minimize potential artifacts (usually between 1 and 10 uM). Generally, 15 minutes to 1 hour is sufficient for cellular uptake and processing of the dyes. |
