• Immunogen: A crude mixture of human urinary proteins precipitated by ammonium sulphate from the urine of a patient suffering from proteinuria.
• Specificity: The antibody MEM-18 reacts with CD14, a 53-55 kDa GPI (glycosylphosphatidylinositol)-linked membrane glycoprotein expressed on monocytes, macrophages and weakly on granulocytes; also expressed by most tissue macrophages. • Application Notes: Western Blot: Non-reducing conditions ELISA: The antibody MEM-18 has been tested as the detection antibody in a sandwich ELISA for analysis of human CD14 in combination with antibody B-A8 (cat. no.NB110-58721). Functional Application: The antibody MEM-18 completely blocks binding of fluorescein (FITC); labeled bacterial LPS to the monocyte surface and it also blocks the binding of CD14 to the extracellular TLR2 domain. This antibody is CyTOF ready.

Background

CD14 is a 53-55 kD glycosylphosphatidylinositol (GPI)-linked membrane glycoprotein also known as LPS receptor. CD14 is expressed at high levels on monocytes and macrophages, and at lower levels on granulocytes. Some dendritic cell populations such as interfollicular dendritic cells, reticular dendritic cells, and Langerhans cells have also been reported to express CD14. As a high-affinity receptor for LPS, CD14 is involved in the clearance of gram-negative pathogens and in the upregulation of adhesion molecules and cytokines expression in monocytes and neutrophils. The M5E2 antibody inhibits monocyte activation and cytokine production induced by LPS.
