200ug/ml of antibody purified from Bioreactor Concentrate by Protein A or G. Prepared in 10 mM PBS with 0.05% BSA & 0.05% azide. Also available WITHOUT BSA & azide at 1.0 mg/ml. (NBP2-33080)

Antibody with azide - store at 2 to 8C. Antibody without azide - store at -20 to -80C.

Specificity
Recognizes a protein of 210-220kDa, which is identified as the complement receptor 1 (CR1)/CD35. This monoclonal antibody is specific for a site in CR1 that is distal from the C3b-binding site, so that it is unable to block CR1 activity. This monoclonal antibody is highly specific to CR1 and shows no cross-reaction with CR2. The primary function of CR1 is to serve as the cellular receptor for C3b and C4b, the most important components of the complement system leading to clearance of foreign macromolecules. The Knops blood group system is a system of antigens located on this protein. Follicular dendritic cells (FDC) are restricted to the B-cell regions of secondary lymphoid follicles. They are CD21+/CD35+/CD1a-. This monoclonal antibody labels follicular dendritic cells and follicular dendritic cell sarcoma.

Application Notes
Use in In-situ Hybridization reported in scientific literature (PMID:34855915).Immunohistochemistry (Formalin-fixed): 1-2ug/ml for 30 minutes at RT. Staining of formalin-fixed tissues requires heating tissue sections in 10mM Tris with 1mM EDTA, pH 9.0, for 45 min at 95C followed by cooling at RT for 20 minutes.

Background

The clone E11 antibody reacts with CD35, also known as complement receptor 1 (CR1), a 210-220 kDa protein (Hogg et al, 1985). CD35 is present on a subpopulation of T lymphocytes (~5%) which are relatively low in CD3 expression. CD35 is also expressed on CD20 positive B lymphocytes, CD14 positive monocytes, and on poly-morphonuclear leukocytes. The primary function of CD35 is to serve as the cellular receptor for C3b and C4b, the most important components of the complement system leading to clearance of foreign macromolecules. The Knops blood group system is a system of antigens located on this protein. In Immunohistochemical staining (IHC) the CD35 antibody strongly labels the follicular dendritic cells (FDCs) and follicular dendritic cell sarcoma. Follicular dendritic cells (FDC) are restricted to the B-cell regions of secondary lymphoid follicles. The antibody is highly specific for CD35 and does not cross react with CD21 (CR2). Furthermore, this CD35 antibody does not block CD35 activity because it recognizes a CD35 epitope distal to the C3b binding site. The antibody has been widely cited in the literature. Examples include its use in antibody panels for immunophenotyping normal bone marrow stem cells by flow cytometry (Laranjeira et al, 2012) and characterizing infections in rheumatoid arthritis patients (Moduko et al, 2012). The CD35 antibody has also been used along with CD23 and CD21 antibodies for identifying FDCs in reactive and neoplastic lymphoid tissue by IHC (Bagdi et al, 2001). Furthermore, the CD35 antibody showed than tonsil-derived mesenchymal progenitor cells acquire an FDC-like phenotype following cytokine stimulation (Cho et al, 2012), and that oncolytic measles viral infection upregulates CD35 (Zhang et al, 2012).
