Background
Clustered Regularly Interspaced Short Palindromic Repeat (CRISPR) is an adaptive immune system in bacteria which provides protection against mobile genetic elements from viruses such as, transposable elements and conjugative plasmids. CRISPR clusters are known to have sequences which are complementary to antecedent mobile elements and target invading nucleic acids, and these sequences are transcribed/processed into CRISPR RNA. CRISPR possess endonuclease activity on pre-crRNA and dsDNA, using different active sites, and in a complexed form, crRNA displaces the non-target DNA strand and guides endo-nucleolytic cleavage of both strands of the DNA. CRISPR associated Cas12a (CRISPR-Cas12a), also known as Cpf1, is emerging as a genome editing tool with different target specificity, when compared to CRISPR-Cas9. CRISPR-Cas12a-assisted recombineering can be used for genetic manipulation of plasmids, plasmid curing and for a more precise selection of target sites for the introduction of desired mutations (point mutations, deletions, insertions, and replacements). In cells subjected to genetic manipulation using this tool, CRISPR-Cas12a antibodies are needed to verify transfection efficiency, CRISPR-Cas12a expression, CRISPR-Cas12a target binding and subcellular localization. Novus Biologicals' CRISPR-Cpf1 antibody (NBP2-62735) is the first-ever commercially launched antibody to meet this need. This antibody is generated against a partial recombinant Francisella tularensis protein (between amino acids 600-850) [NCBI WP_003034647.1], and validated for Western blot using a partial recombinant CRISPR-Cpf1 protein (12kDa), making it a great tool for a direct detection of CRISPR-Cpf1/CRISPR-Cas12a complex (at protein level) superior to the use of epitope-tag based validation approaches.
