In Western blot multiple bands are seen around 45 and 100 kDa (dimer) in SF9 cell lysates overexpressing D1a and a single band at 90 kDa in striatal tissue lysate. Do NOT boil samples.

Background
Dopamine is a unique brain neurotransmitter which is responsible for the regulation of versatile processes such as motor activity, cognition, motivation, emotion, food intake, endocrine secretions etc. and dysfunctions in dopaminergic systems have been linked to drug addiction, schizophrenia, Parkinson's disease, Tourette's syndrome, attention-deficit and hyperactivity disorders as well as hyperprolactinemia. Dopamine Receptor D1 (DRD1), a GPCR protein, is the D1 subtype of dopamine receptors (DR) which represents the most abundant DR in CNS with capability of stimulating adenylyl cyclase and activating cyclic AMP-dependent protein kinases. DRD1 localize to cell membrane/ER membrane as multi-pass membrane protein and its transport from ER to cell surface is regulated by interaction with DNAJC14. DRD1 is highly expressed in the brain tissues, especially in the striatum, the nucleus accumbens, and the olfactory tubercle. DRD1 regulate neuronal growth and development, mediate behavioral responses, and modulate dopamine receptor D2-mediated events.

I am writing a manuscript and referring to results we obtained with your Dopamine Receptor D1 antibody. (These are results listed on your website: Journal of Neuroscience 29: 15001.) Your datasheet states that the immunogen was 123 residues of the rat Dopamine Receptor D1a. I realize the amino acid sequence that you used is proprietary, and do not need to know which residues you used. However, can you tell me the position of some of the amino acids so that I can state, with certainty, if these residues are not in the rat Dopamine Receptor D1b amino acid sequence?
I have spoken with our lab manager, and after pulling the original production data for this antibody, it does appear that the protein sequence used to synthesize the antibody was in fact the last 123 amino acids from residues 323 to 446 at the C-terminal. This updated immunogen information has been added to the datasheet to reflect that the antibody targets the C-terminus. Keep in mind the protein used contained to synthesize the antibody contained a HIS tag. I hope this information is helpful in your publication, and we are happy to hear that you are using our product for your published data.
