Application Notes
This mAb is a very useful tool for clinical and experimental allergy studies. BMK-13 is a pan-eosinophil marker in both frozen sections and paraffin embedded tissues. It stains in bronchial sections 98% of the periphere blood eosinophils. Further, the antibody is most useful for immunohistochemistry on frozen sections is the APAAP method. BMK-13 also binds to eosinophils in paraffin-embedded tissue. A 1:30 dilution in PBS is recommended. Incubation time: 1 hour at RT. Enzymatic pretreatment is necessary. This antibody is suitable for blood smears (cytospins) if fixed with methanol-acetone mixture (1:1, v/v). It is unlikely, without permeabilization, that the BMK-13 is suitable for the flow cytometry because BMK-13 stains intracellular, stored proteins.

Background

PRG2 is a cytotoxin and helminthotoxin. Also induces noncytolytic histamine release from human basophils. Involved in antiparasitic defense mechanisms and immune hypersensitivity reactions. The proform acts as a proteinase inhibitor, reducing the activity of PAPPA. In pregnancy serum, the proform exists as a disulfide-linked 2:2 heterotetramer with PAPPA, as a disulfide-linked 2:2 heterotetramer with AGT, and as a complex (probably a 2:2:2 heterohexamer) with AGT and C3dg.
