The HER2/LUCPorter reporter cell line is designed to monitor the induction of HER2 and can be used for screening of agonists, antagonists or signaling inhibitors of HER2 induction as well as for studying the HER2 induction-related signaling pathways.

Complete Growth Medium
DMEM with 4.5 g/L glucose + 10% FBS + 4 mM L-glutamine + 1 mM sodium pyruvate + 100 units/ml penicillin + 0.1 mg/ml streptomycin + 3 ug/ml puromycin.

Safety Considerations
Assume all cultures are hazardous since they may harbor latent viruses or other organisms that are uncharacterized. The following safety precautions should be observed. - Use pipette aids to prevent ingestion and keep aerosols down to a minimum. - No eating, drinking or smoking while handling the reporter line. -Wash hands after handling the reporter cell line and before leaving the lab. - Decontaminate work surface with disinfectant or 70% ethanol before and after working with the reporter line. -All waste should be considered hazardous. - Dispose of all liquid waste after each experiment and treat with bleach.

Background

As a member of the epidermal growth factor receptor family, HER2 (human epidermal growth factor receptor 2; also known as Neu, ErbB-2, CD340 or p185) has been known to play an important role in breast cancer development and progression. Dysregulation of HER2 signaling such as excess gene amplification and overexpression may result in deleterious effects of HER2 in tumor cells. Elevation of HER2 is often observed in a high proportion of metastatic breast cancers, enhancing aggressive tumor cell growth. Hence therapeutic inhibition of HER2 in metastatic breast cancers would suppress tumor cell proliferation and provide synergistic effects with other cancer treatments. The HER2/LUCPorter reporter cell line is a semi-constitutively active cell line and also ligand/agonist-inducible cell line as shown in Figures 1 and 2.
