Western Blot: Exosome Detection (Western Blot) Antibody Pack [NBP3-11740] - Isolation and characterization of exosomes from cell culture supernatant and human blood. Immunoblot of exosomal markers CD81 (~30 kDa) using CD81 Antibody (NB100-65805), CD63 (~55 kDa), and LAMP2B (~50 kDa) in exosomes (5 ug lysates) isolated from the serum of breast cancer patients and healthy control. Grp94 (~100 kDa) and beta-actin (~42 kDa) serve as a negative control and a loading control, respectively. Western Blot: Exosome Detection (Western Blot) Antibody Pack [NBP3-11740] - Western blot shows lysates of TIME human endothelial cell line and human platelets. PVDF membrane was probed with 2 ug/mL of Mouse Anti-Human CD9 Monoclonal Antibody (Catalog # MAB25292) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # HAF018). A specific band was detected for CD9 at approximately 24 kDa (as indicated).

Exosome Detection (Western Blot) Antibody Pack [NBP3-11740] - Western blot shows lysate of human liver tissue. PVDF membrane was probed with 1 ug/mL of Mouse Anti-Human Serum Albumin Monoclonal Antibody (Catalog # MAB1455) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # HAF018). A specific band was detected for Albumin at approximately 65-70 kDa (as indicated).

Background

The Exosome Detection (Western Blot) Antibody Pack contains antibodies from each category recommended by the International Society for Extracellular Vesicles (ISEV) for characterization of extracellular vesicles (EVs), with data on exosomes published in peer-reviewed journals. It includes three tetraspanins (CD9, CD63, and CD81), one intracellular protein associated with multivesicular body (MVB) formation (TSG101), and one chaperone protein (HSP70). The pack also contains two antibodies to determine the degree of contamination of exosome prep with secretory vesicles (Calnexin) and circulating free protein (albumin). All antibodies have been validation for use in western blotting.
