Background

FKBP1B, also known as peptidyl-prolyl cis-trans isomerase FKBP1B, is a member of the immunophilin protein family which plays a role in immunoregulation and basic cellular processes involving protein folding and trafficking. This protein is a cis-trans prolyl isomerase that binds the immunosuppressants FK506 (tacrolimus) and rapamycin (sirolimus). It is highly similar to the FK506-binding protein 1A. Its physiological role is thought to be in excitation-contraction coupling in cardiac muscle. Recombinant human FKBP1B protein, fused to His-tag at N-terminus, was expressed in E.coli and purified by using conventional chromatography.

    • The molecular weight for NBP1-72505 is 14.2 kDa (not including the His tag ~26kDa) and the concentration is 0.25mg/mL.
  2. I was wondering whether the product is suitable for any kind of assay/experiment since the only use suggested is PAGE?
    • To answer your question regarding the applications FKBP1B Protein (NBP1-72505) can be used for, it has already been tested for its ability to cleave suc-AAFP-pNA but our lab suspects it can be used in other functional assays, although it hasn't been confirmed yet. For untested applications we recommend buying smaller amounts first.
  3. For NBP1-72505 (FKBP12.6) for immuno-assay, is the 6HIS ist bound to the C-end or the N-end?
    • The His-tag is linked at the N-t of the protein. I copied the a.a. sequence of FKBP12.6 as follow: "MGSSHHHHHH SSGLVPRGSH RSMGVEIETI SPGDGRTFPK KGQTCVVHYT GMLQNGKKFD SSRDRNKPFK FRIGKQEVIK GFEEGAAQMS LGQRAKLTCT PDVAYGATGH PGVIPPNATL IFDVELLNLE", where the six His within the first 10 a.a. is the His-tag.
