Human cerebral cortical neurons are derived from NIH approved induced pluripotent stem cell (iPSC) lines under a fully defined neural induction condition (Shi, Y. et al, 2012a, b, c). The hCCNs & hyCCNs express typi...cal markers of cerebral cortical neurons such as Tbr1, Ctip2, Brn2 and Cux1. The cortical neurons are electrically active and have the ability to form functional synapses and circuits in vitro. In addition, the cortical neurons transplanted into the neonatal rat cortices can project long distances to innervate other regions of cerebral cortex. Cerebral cortical neurons have been implicated in numerous neurological diseases including Alzheimer's disease, autism, epilepsy and stroke. Despite strong interest in these diseases, high quality human in vitro cell models for cortical neurophysiology were not available until now.
