Assay 1: Fully biologically active when compared to standard. Measured in a serum-free cell proliferation assay using human MCF-7 cells. Background
Insulin-like Growth Factor I (IGF-I), also known as Somatomedin C, is the dominant effector of Growth Hormone (GH). IGF-I regulates glucose, fatty acid, and protein metabolism, steroid hormone activity, cartilage and bone metabolism, muscle regeneration, tumor progression, and cellular differentiation and migration. IGF-I effects are mediated primarily by IGF-I R, and its bioavailability is regulated by IGF binding proteins (IGFBPs). Mature human IGF-I shares about 95% aa sequence identity with the mouse and rat IGF-I. Long R3 IGF-I (LR3 IGF-I) is a synthetic analog of IGF-I that has a Glu3Arg substitution and a 13 aa N-terminal extension derived from methionyl porcine GH. It retains the ability to bind IGF-I R and Insulin R but shows reduced affinity for IGFBPs. As a result, LR3 IGF-I has an increased half-life and displays increased biological potency compared to IGF-I.
