Background

IL17, also known as CTLA8, is a T cell derived hematopoietic cytokine. It was originally cloned from a T cell hybridoma produced by fusion of a mouse cytotoxic T cell clone and a rat T lymphoma. IL17 exhibits multiple biological activities on a variety of cells including: the induction of IL6, IL8 and GCSF production in fibroblasts; the enhancement of surface expression of ICAM 1 in fibroblasts; activation of NF kappaB and costimulation of T cell proliferation. IL17 is an approximately 16 kDa polypeptide of 136 amino acids. The precursor form of IL17 consists of 155 amino acids. To generate the mature IL17 (136 amino acids), the precursor cleaves a 19 amino acid signal peptide. Human IL17 shows approximately 62.5% amino acid homology to mouse IL17 and 58% amino acid homology to rat IL17.

  IL-17/IL-17A antibody used for IHC-P on the AD lesions from hyperbaric oxygen therapy (HBOT) or applying an oxygen-carrying chemical, perfluorodecalin (PFD) treated murine model of AD that was developed by repeated application of a chemical irritant (1% 2,4-dinitrochlorobenzene) and house dust mite (Dermatophagoide farinae) extract on one ear of BALB/c mice. Figure 6 shows a decreased IL-17A expression in the AD lesions of HBOT or PFD treated mice.

1. I want to stain some T cell subsets (Th17 and Th1), do you have antibodies for that? I need for flow cytometer in sheep or goat.
   • Unfortunately, we have neither a CD4 antibody nor an IL-17 antibody that has been validated in sheep or goat, although if you would like to try one you would again be eligible for our Innovators Reward Program. 2. I'm looking to establish an ELISA for IL-17 and want your recommendation on which antibody pairs to buy. Least expensive ones, please. Also, I'd like the detection antibody to be fluorescently labeled.
   • Thank you for your patience while I reviewed our ELISA validated products for IL17. I was not sure what species you were going to be investigating, so I looked up our data for the human protein. If this is incorrect, just let me know and I can look up products for the species you will be working with. As for antibody pairs, we only had one set that have been tested together: Capture: NBP2-22531 Detection: NBP2-22532. The detection product is only available currently with a biotin conjugate. We can however offer custom conjugation for this product to any of our available conjugates. Please see the this link to see the available fluors. The typical fee for custom conjugation is usually about $80 additional. Once you know which fluor you would like, I can provide you with a quote. We do also offer several ELISA kits for this target (they typically employ HRP or Biotin detection systems, so I am not sure if they will work for your needs).
3. I would like to know if the IL17 Antibody (NBP1-72027) has been evaluated for different mice strains, and, if not, which mouse strain was it designed for?
   • This antibody was designed to work in all mouse strains. The IHC image on the product datasheet came from the spleen of a Swiss-Webster mouse specifically.
