Description |
The TNF beta Prom/LUCPorter™ reporter cell line can be used for screening of agonists, antagonists or signaling inhibitors related with TLR signaling pathways as well as for studying the TNF beta induction-related signaling pathways.
TNF beta is one of the major proinflammatory cytokines, which is produced predominantly by mitogen-stimulated T lymphocytes and leukocytes. TNF beta is involved in the regulation of various biological processes such as cell proliferation, differentiation and apoptosis. TNF beta gene polymorphisms are also significantly associated with disease susceptibility and phenotype in patients with psoriatic arthritis. The TNF beta induction by various Toll-like receptor ligands is shown in Figure 1. In the test, all Toll-like receptor (TLR) ligands except TLR5 ligand significantly mediated activation of the TNF beta promoter which was quantified by luciferase activity (figure 1).

Notes
Assume all cultures are hazardous since they may harbor latent viruses or other organisms that are uncharacterized. The following safety precautions should be observed.

- Wash hands after handling the TNF beta reporter cell line and before leaving the lab.
- Decontaminate work surface with disinfectant or 70% ethanol before and after working with the cell line.
- All waste should be considered hazardous.
- Dispose of all liquid waste after each experiment and treat with bleach

Background
TNF-beta (tumor necrosis factor beta, lymphotoxin alpha, TNFSF1, TNF ligand superfamily member 1) a member of the TNF ligand superfamily and a potent lymphoid factor, is mainly produced by activated T and B lymphocytes and leukocytes. TNF-beta is also produced by fibroblasts, astrocytes, myeloma cells, endothelial cells and epithelial cells. TNF-beta mediates inflammatory and immune responses, has cytotoxic activity on tumor cells and other target cells and shares many of the same bioactivities as TNF alpha, including regulation of cell proliferation, differentiation and apoptosis. TNF-beta is not species-specific and human TNF-beta is active on murine cells with a slightly reduced specific activity.
