Background

Principal Names: Neuregulin 1 Official Gene Symbol: Nrg1 Gene ID: 112400 (Rat) Gene Map Locus: 16q12.3 (Rat). Heregulin or Neu differentiation factor (NDF), a 44-kD polypeptide, is a member of the neuregulin family that were first isolated as ligands for the HER2 (ergB2, or p185neu) receptor protein tyrosine kinase. It is a transmembrane protein whose extracellular portion contains an EGF-like domain that probably functions as a receptor recognition site. In addition, the ectodomain contains one immunoglobulin homology unit. It activates ErbB2 via its heterodimerization with ErbB3 or ErbB4. Eight differentially spliced isoforms of the gene exists. All NDF isoforms encode membrane-anchored precursor proteins from which the mature growth factor is proteolytically released. The protein acts as a regulator of growth and differentiation of colonic epithelial cells glial, neuronal, and skeletal muscle cells, cause apoptosis when overexpressed in tissue culture cells, mediate proliferation, differentiation and gene expression in human schwann cells, induces expression of acetylcholine receptor in synaptic vessicles during the formation of the neuromuscular junction, stimulates lobuloalveolar budding and milk production in the mammary gland and induces differentiation of mammary tumor cells.
