This antibody reacts with human PTEN also known as MMAC or TEP1. PTEN is a tumor suppressor gene, which encodes a multifunctional phosphatase and which is expressed almost ubiquitously and regulates the cell cycle, apoptosis, and cell adhesion. Deletions and mutations of PTEN occur in a range of cancers including breast cancer, malignant melanoma, endometrial carcinoma, bladder carcinoma, small cell lung carcinoma, and endometrial ovarian carcinoma.

Background

The PTEN tumor suppressor gene acts as a phospholipid phosphatase. Under normal growth conditions, stimulatory signals from the insulin receptor activate the enzyme PI3-K, which phosphorylates phosphotidylinositol 4,5-bisphosphate (PIP2) to generate phosphotidylinositol 3,4,5-triphosphate (PIP3), a lipid molecule. Downstream, PIP3 activates several effectors, including the proto-oncogene product PKB/Akt. The role of PTEN is to dephosphorylate PIP3, acting as a negative control on PKB/Akt activation. If a mutation in PTEN causes it unable to carry out its phosphatase function, PIP3 can no longer be deactivated, so it continues to propagate its signal downstream. This may result in the continued activation of PKB/Akt, which, in combination with other factors, could lead to increased cell growth and possible tumor development.

    • Yes, all of these antibodies are useful for IHC-P with DAB based detection. Please note that the PTEN Antibody (28H6) NB200-175 has been discontinued, however.
