Application Notes
Use in Immunohistochemistry-paraffin reported in scientific literature (PMID: 30015619). This SC35 Antibody (SC-35) is validated for IHC-Fr and IHC-Free Floating from a verified customer review. The antibody labels SC-35 as a speckled pattern in the nucleoplasm, excluding the nucleoli. Other uses include inhibition and depletion of splicing activity in nuclear extracts and immunoaffinity purification. IHC-P: See Nie et al 2002 paper where it appears no antigen retrieval is necessary. Block in normal serum 10% in TBS (pH 7.6) for 30min then incubate antibody overnight at 4C. Use ABC amplification- DAB detection. WB: Detects a doublet of approximately 35 kDa. *Optimal dilutions/concentrations should be determined by the end user.

Background

SC35 is involved in pre-mRNA splicing and is found in the bodies in the nucleus referred to as speckles, SC 35 domains or splicing factor compartments (SFCs). The SC-35 clone is frequently used as a marker for these bodies, the role of which is controversial. Speckles correspond largely if not entirely to ultrastructures termed interchromatin granule clusters (IGCs). SC35 is excluded from the nucleolus.
