Notes
Unprocessed SREBP1 is an ~122 kDa integral membrane protein that moves from ER to golgi for processing. The first cleavage is performed by S2P at residue 490, and the resulting active portion is then translocated out of the golgi. This is the ~65 kDa mature form that is detected by NB100-2215. The second cleavage is performed by S1P at residue 530, and the resulting C-term portion (from residues 530-1147) remains in the golgi where it is detected as a ~65 kDa protein by NB100-60545.

SREBP1 (sterol regulatory element binding protein 1, also called SREBF1 or BHLHD1) is a bHLH-LZ (basic helix-loop-helix-leucine zipper) transcription factor that acts as a key lipogenic transcription factor controlled by glucose/insulin and preferentially regulates lipogenesis via activating genes involved in fatty acid as well as triglyceride synthesis. It is a transcriptional activator required for lipid homeostasis and regulates transcription of the LDL receptor gene as well as the fatty acid and to a lesser degree the cholesterol synthesis pathway. SREBP1 has a dual sequence specificity binding to both an E-box motif (5'-ATCACGTGA-3') and to SRE-1 (sterol regulatory element 1; 5'-ATCACCCCAC-3'). It exists as multi-pass membrane protein in the membranes of ER, Golgi apparatus and cytoplasmic vesicle /COPII-coated vesicles and moves from ER to Golgi in the absence of sterols. It forms a tight complex with SCAP in ER membranes and interacts with LMNA. AMPK phosphorylate SREBP1 leading to suppression of protein processing and nuclear translocation, and target gene expression repression. Moreover, SIK1 mediated phosphorylation at Ser-402 site represses its activity through inhibition of DNA-binding.

FAQs for SREBP1 Antibody (NB100-60545). (Showing 1 - 2 of 2 FAQs).
I am looking for SREBF1 antibodies that can cross-react with zebrafish srebf1. I am wondering whether you have any SREBF1 antibodies that have been tested for zebrafish or have high sequence identity in the immunogen region.
Unfortunately we do not have any antibodies that have been tested in zebrafish. I have included the immunogen sequence for the antibodies it might be worth considering. I am not sure exactly what the zebrafish sequence is, I was unable to find it on uniprot.org, so I was unable to check the homology. The following antibodies all have available sample sizes. If you decide to try one you would qualify for our Innovators Reward Program. NB600-582 - 301-407 aa, NB100-2215 - 300-400 aa, NB100-60545 - 700-800 aa.
A customer is interested in NB100-60545 for use with zebra fish. Would you be able to blast the exact immunogen sequence to see what the homology is?
The homology for the immunogen of this antibody is approximately 78%.
SREBP: Gatekeeper of Cholesterol Homeostasis SREBP1 (sterol-regulatory-element-binding protein 2) is a basic-helix-loop-helix-leucine zipper (bHLH-ZIP) transcription factor. It regulates sterol and cholesterol homeostasis by controlling enzymes involved in cholesterol synthesis and uptake, e.g. ... Read full blog post.
