Streptavidin Antibody

Western Blot: Streptavidin Antibody [NB100-1675] - Lane 1: Opal Prestained Molecular Weight Ladder. Lane 2: Streptavidin Load: 50ng. Primary Antibody: Rabbit Anti-Streptavidin at 1:1000 overnight at 4C. Secondary Antibody: Goat anti-Rabbit HRP at 1:70,000 for 30min at RT. Blocking: BlockOut for 30 min at RT.

Western Blot: Streptavidin Antibody [NB100-1675] - Streptavidin antibody and Biotin conjugated Rabbit anti-trypsin inhibitor antibody were used to detect target proteins Trypsin Inhibitor (left) and Streptavidin (right) under reducing (R) and non-reducing (NR) conditions. Reduced samples of purified target proteins contained 4% BME and were boiled for 5 minutes. Samples of ~1ug of protein per lane were run by SDS-PAGE. Protein was transferred to nitrocellulose and probed with 1:1000 dilution of primary antibody (ON 4 C). Detection shown was using Dylight 649 conjugated Donkey anti rabbit ( 1:10K 1.5 hr RT) and imaged on the BioRad VersaDoc System.

Streptavidin Antibody [NB100-1675] - A three-fold serial dilution of DyLight(TM) 488 Conjugated Streptavidin starting at 200ng was spotted onto 0.45 um nitrocellulose and blocked in 1% BSA-TTBS (diluted to 1X) 30 min at 20C. Streptavidin antibody was incubated in Blocking Buffer for Fluorescent Western Blotting at 1:1,000 for 1 hr at 20C. An HRP Goat anti-Rabbit secondary antibody was incubated at 1:40,000 for 30 min at 20C and imaged using the Bio-Rad VersaDoc(R) 4000 MP.

This antibody is an IgG fraction antibody purified from monospecific antiserum by a multi-step process which includes delipidation, salt fractionation and ion exchange chromatography followed by extensive dialysis against the buffer stated above. Assay by immunoelectrophoresis resulted in a single precipitin arc against anti-Rabbit Serum as well as purified and partially purified Streptavidin [Streptomyces avidinii].]

For extended storage aliquot contents and freeze at -20C or below. Avoid cycles of freezing and thawing. Centrifuge product if not completely clear after standing at room temperature. This product is stable for several weeks at 4C as an undiluted liquid. Dilute only prior to immediate use.

Background

Streptavidin is a tetrameric protein purified from Streptomyces sp. that binds very tightly to the vitamin biotin with a Kd of ~ 10-14 mol/l. The high affinity recognition of biotin and biotinylated molecules has made streptavidin one of the most important components in diagnostics and laboratory kits.
