In vitro assay: Thunder-Link (R) PLUS oligo Antibody Labeling Kit [425-0000] - Immuno-qPCR data generated using Thunder-Link. A mouse monoclonal antibody specific for human CRP (clone C7) was purchased in unconjugated format from HyTest. The antibody was conjugated to an oligonucleotide using a Thunder-Link kit, and was used as a detection antibody in a sandwich immuno-PCR assay with a polyclonal anti-CRP antibody as the capture reagent. The graph shows a plot of the number of PCR cycles undertaken vs. fluorescence intensity generated by SYBR Green containing PCR probes at particular antigen concentrations.

In vitro assay: Thunder-Link (R) PLUS oligo Antibody Labeling Kit [425-0000] - The graph shows this data following conversion to antigen amount vs. cycle number to enable the calculation of a standard curve. The results show that the assay utilizes 1000-fold less capture antibody and 100-fold less detection antibody, and provides 1000-fold greater sensitivity than the equivalent ELISA.

• Application Notes: Warm the Thunder-Link PLUS Conjugate Clean Up Reagent by placing the tube in warm water (not warmer than 40C) for 10 minutes, then shaking to re-dissolve the contents. Once dissolved, maintain the tube at ~22C to prevent any crystal formation before use.

1. If I want to conjugate an oligo to my antibody using your kit (Thunder-link Plus kit), what type of modification needs to be in my oligo.  ..show answer..
   • The oligo must be between 10 and 120 base pairs in length and contain a terminal amine group, which must be added during synthesis. All commercial oligo suppliers offer this modification. The efficiency of conjugation is slightly higher with 5’aminated oligos.
2. We recently ordered a Thunder-Link PLUS oliogo Antibody Conjugation Kit to tag some of our commercial antibodies. The protocol specifies that purified antibodies must be at a concentration of 1mg/mL, as the Activation reagents are designed to activate 100ug of antibody in 100uL reaction volume. Our antibodies are too dilute, at 0.5mg/mL commercial concentration. The protocol suggests contacting technical support. What are your recommendations?  ..show answer..
   • We have 2 products designed for antibody concentration, clean up and changing the buffer the antibody is in. These products are Antibody concentration and clean up kit (262-0010) and Antibody concentration and clean up kit (861-0010). But you can use a simple spin column with a 50-100kDa cut off. We do not have these in our portfolio but they are available from various companies e.g Millipore, ThermoFisher, Pierce, Bio-Rad etc. This is very simple and inexpensive way to concentrate your antibody.
