Western Blot: YTHD1 Antibody [NBP2-76361] - Samples: Whole cell lysate (50 ug) from GaMG, HEK293T, mouse TCMK-1, and mouse NIH 3T3 cells prepared using NETN lysis buffer. Antibody: Affinity purified rabbit anti-YTHDF1 antibody NBP2-76361 used for WB at 1:1000. Detection: Chemiluminescence with an exposure time of 30 seconds.

Immunoprecipitation: YTHD1 Antibody [NBP2-76361] - Samples: Whole cell lysate (1.0 mg per IP reaction; 20% of IP loaded) from GaMG cells prepared using NETN lysis buffer. Antibodies: Affinity purified rabbit anti-YTHDF1 antibody NBP2-76361 used for IP at 100 ul per reaction. YTHDF1 was also immunoprecipitated by rabbit anti-YTHDF1 antibody NBP2-76360. For blotting immunoprecipitated YTHDF1 was used at 1:1000. Detection: Chemiluminescence with an exposure time of 30 seconds.

Background

YTHDF1 specifically recognizes and binds N6-methyladenosine (m6A)-containing mRNAs, and promotes mRNA translation efficiency. M6A is a modification present at internal sites of mRNAs and some non-coding RNAs and plays a role in the efficiency of mRNA splicing, processing and stability. Acts as a regulator of mRNA translation efficiency: promotes ribosome loading to m6A-containing mRNAs and interacts with translation initiation factors eIF3 (EIF3A or EIF3B) to facilitate translation initiation.
