Mouse NFATC2/NFAT1 Luciferase - (LUCPorter™) Stable Reporter Cell Line Summary

Description
The NFAT / LUCPorter™ reporter cell line can be used for screening of agonists, antagonists or signaling inhibitors related with the NFAT-involved signaling pathways. so that the cell line is designed to measure the transcriptional activity of NFAT. NFAT is a transcription factor originally found in activated T lymphocytes, and is now known to regulate not only T cell activation and differentiation but also the function of other immune cells including dendritic cells, B cells and megakaryocytes.

Numerous studies have shown that aberrant NFAT signaling related with overexpression and hyperactivity is associated with human cancers including breast cancer, prostate cancer, pancreatic cancer and B cell lymphomas. The NFAT induction by calcium ionophore A23187 and/or phorbol 12-myristate 13-acetate (PMA) is shown in Figures 1 and 2. In the test, calcium ionophore induced the transcriptional activity of NFAT in a dose response manner (Figure 1), and the activity was further enhanced by PMA addition (Figure 2).

Reconstitution Instructions
Complete Growth Medium: DMEM with 4.5 g/L glucose + 10% FBS + 4 mM L-glutamine + 1 mM sodium pyruvate + 100 units/ml penicillin + 0.1 mg/ml streptomycin + 3 ug/ml puromycin.

Notes

Assume all cultures are hazardous since they may harbor latent viruses or other organisms that are uncharacterized. The following safety precautions should be observed.
- Use pipette aids to prevent ingestion and keep aerosols down to a minimum.
- No eating, drinking or smoking while handling the NFAT reporter line.
- Wash hands after handling the NFAT reporter cell line and before leaving the lab.
- Decontaminate work surface with disinfectant or 70% ethanol before and after working with the reporter line.
- All waste should be considered hazardous.
- Dispose of all liquid waste after each experiment and treat with bleach.

Background

The nuclear factor of activated T-cells (NFAT) transcription complex is required for the expression of a group of proteins that collectively regulate the immune response. Four NFAT proteins, encoded on separate genes and expressed as several splice variants, have been described: NFAT1, NFAT2, NFAT3, and NFAT4. These proteins show a low level of sequence similarity with the Dorsal/Rel/NFkB family of transcription factors. Another NFAT-related protein termed NFAT5 differs from isoforms 1-4 in that it lacks many of the Fos/Jun contact sites observed in its predecessors and its subcellular localization is not calcineurin-dependent.
