Western Blot: NUT Antibody (46B3D2.1G6) [NBP2-31146] - WB analysis of NUT/ NUTM1 protein in (A) human testis lysate and on (B) partial recombinant NUT protein (expected molecular weight ~17kda) with NUT antibody (clone 46B3D2.1G6) at a concentration of 2 ug/ml. In human testis, this antibody detected Isoform 1 and Isoform 2 at ~120 kDa and ~46kDa respectively.

Immunocytochemistry/Immunofluorescence: NUT Antibody (46B3D2.1G6) [NBP2-31146] - NUT antibody was tested in A431 cells with Dylight 488 (green). Nuclei and alpha-tubulin were counterstained with DAPI (blue) and Dylight 550 (red). An antibody concentration of 0.1 ug/ml was used. Image objective 40x.

Background

NUT is known to fuse with BRD3 and BRD4 in NUT midline carcinoma (NMC), a rare and aggressive human cancer. In the majority of NMCs (~75%), most of the coding sequence is fused with BRD4 creating chimeric genes that encode BRD-NUT fusion proteins. In other cases, it fuses with BRD3 or an unknown partner gene. This tumor is often termed NUT-variant. To date, NMCs are still frequently undiagnosed or misdiagnosed and there are no effective treatment options. [PMID: 20951314]
