Description
The TEAD/LUCPoter(TM) reporter cell line is designed to monitor the induction of TEAD and can be used for screening of agonists, antagonists or signaling inhibitors of TEAD induction as well as for studying the TEAD induction-related signaling pathways.

The TEAD/LUCPorter(TM) reporter cell line is a stably transfected HEK 293 cell line which expresses an optimized Renilla luciferase reporter gene (RenSP) under the transcriptional control of the human transcription enhancer factor TEF-1 (TEAD1) promoter. TEAD1 is one of the four human homologous members of the TEAD family which are transcriptional mediators of Hippo signaling pathway and play a crucial role in development, regulating cell proliferation and contact inhibition. TEAD1 is also known to facilitate the expression of cardiac-specific genes which are important for the differentiation of cardiac muscle. Since the Hippo pathway can alter gene expression in response to changes in cell shape, adhesion and density, its dysregulation may influence the metastatic cascade of cancerous cells. Recent studies have indicated that the metastatic potential of breast cancer and melanoma cells is correlated with increased TEAD transcriptional activity which is mediated through the association of TEAD and its transcriptional coactivator Yes-associated protein (YAP). The TEAD/LUCPorter(TM) reporter cell line, which can be used as a tool for identification of TEAD1 induction modulators, is an inducible reporter cell line. This cell line can be induced by treatment of phorbol 12-myristate 13-acetate (PMA) as shown in Figure 1.

Complete Growth Medium: DMEM with 4.5 g/L glucose + 10% FBS + 4 mM L-glutamine + 1 mM sodium pyruvate + 100 units/ml penicillin + 0.1 mg/ml streptomycin (Note: The selection agent for this cell line is puromycin at 3 ug/ml).

Background

This gene encodes a ubiquitous transcriptional enhancer factor that is a member of the TEA/ATTS domain family. This protein directs the transactivation of a wide variety of genes and, in placental cells, also acts as a transcriptional repressor. Mutations in this gene cause Sveinsson's chorioretinal atrophy. Additional transcript variants have been described but their full-length natures have not been experimentally verified.
