We would like to know about the staining pattern of NBP2-15177. I checked Uniprot and found ZNF639 shall be expressed at nucleus, but later I found it also presents at cytoplasm based on the info from http://www.proteinatlas.org/ENSG00000121864/tissue.
- It is a relatively new and less published target, and it will be too early to make any generalization on the cellular localization of ZNF639 protein. Imoto et al. 2003 (Identification of ZASC1 encoding a Kruppel-like zinc finger protein as a novel target for 3q26 amplification in esophageal squamous cell carcinomas.) performed their studies on transfected cells and found the overexpressed protein to be nuclear, however, on page 5693, column 2, last sentence of paragraph 1 of their research paper, they wrote that : " although the subcellular localization should be confirmed by investigating the presence of endogenous ZASC1 using specific antibody, our result suggested that ZASC1 might function as a nuclear transcription factor". Bogaerts et al.2005 (Nuclear translocation of alphaN-catenin by the novel zinc finger transcriptional repressor ZASC1.) tested the subcellular localization of the endogenous ZASC1 protein in MCF7/AZ cells with monoclonal antibody 8D4/1 and reported it as a nuclear target. However, from the images shown in this article, it looks like besides strong nuclear expression, this target shows mild cytoplasmic expression also (endogenous as well as the overexpression situations)! Human Protein Atlas team tested ZASC1 in 81 normal tissue/cell types, and they found a positive signal in 45 of the tested sample types! They employed two different antibodies (specificity tested with WB and protein array), and found nuclear as well as cytoplasmic staining. In our testing with #NBP2-15177 in HBL435 xenograft, we observed strong nuclear with mild cytoplasmic staining.
